General Information Anti-Nuclear Antibodies (ANA) are autoantibodies which binds to cellular nuclear antigens including ds-DNA, ss-DNA histones, ribonucleoproteins (RNP) and the SS-A, SS-B, Sm antigens, etc. The presence of ANA is a classic marker of systemic rheumatic diseases including systemic lupus erythematosus (SLE), Sjogren's Syndrome (SS), Mixed connective tissue diseases (MCTD), and progressive systemic sclerosis. A great deal of our understanding of autoimmune diseases comes form the study of animal models that mimic the clinical picture in humans. Anti-ds-DNA and Anti-Sm appear to occur only in SLE, while others (ss-DNA, histones, nRNP, SSA/Ro, and SSB/La) occur in other conditions as well. The presence of ss-DNA antibody is frequently associated with drug-induced autoimmunity. Anti-histone antibodies are commonly found in drug induced lupus (90-95%) than SLE (20-30%). The most commonly studied mouse models are NZB/NZW, MRL/lpr and the NZW/SWR mice. The NZW and SWR mice are clinically and Immunologically normal. NZB stain, however, has a variety of immunological abnormalities including ANAs but can live to about 18 month. The NZB/NZW or NZB/SWR F1 (SNF1) hybrid mice, by contrast, develop complex autoimmune diseases with death in about 8 month of age. They also have high titers of ANAs. ADI?s ssDNA ELISA, provides semi-quantitative assay to detect the presence of ssDNA antibody in mouse serum. This kit does not measure total mouse IgG or other serum proteins. ADI has other ELISA kits for mouse serum IgG, IgA, IgM, IgE etc (consult web site for details).

SpCrossreactivity Serum samples known to be positive for anti-extractable nuclear antibodies (ENA), anti-dsDNA, anti-rheumatoid factor (RF), anti-toxoplasma gondii IgG, IgM, and anti-Cytomegalovirus IgG were tested in anti-DNA ELISA kit were found to be negative, confirming the specificity of this assay. The S1-nuclease treatment of coated plates does not affect the binding of anti-DNA IgG indicating virtual absence of ssDNA on the coated surface. The antibody conjugate used in the kit is anti-mouse IgG (H+L) that will detect most Ig?s (IgG+A+M) antibodies to DNA. ADI has developed other kits for the measure of anti-ssDN..
