Inhibitor of HSP90. Novobiocin interacts with the C-terminal ATP-binding domain of HSP90, unlike 17-AAG (A1256), Geldanamycin (G1047), Herbimycin A (H1050), or Radicicol (R1130) which all bind to the N-terminal ATP-binding site of HSP90. Also useful in production of positively supercoiled plasma DNA, targeting the nucleotide-binding site of gyrase B.
